Chemical biology, synthetic biology and multi-omics tools to read, write and repair the genome.
We develop chemical-biology and synthetic-biology tools to interrogate how site-specific histone modifications shape chromatin structure and gene regulation in living cells. By combining genetically encoded photoaffinity probes, crosslinking proteomics and genome-wide sequencing, we built the SiTomics platform — a single-site-resolved multi-omics strategy that links the chromatinized proteome to the genome, and revealed the central role of super-enhancers in the metabolite–modification–transcription axis.
We design precise genome- and cell-based therapies that target the root genetic cause of inherited disorders. Our recent work applied adenine base editing to correct the most common PEX1 variant causing Zellweger spectrum disorder, restoring peroxisome and liver function in vivo and in patient-derived cells, and provided the deaminase that was subsequently used in a first-in-human personalised gene-editing treatment.
How do repetitive elements and transposable elements shape genome architecture and regulatory innovation across species? We combine comparative genomics, AI-assisted sequence analysis and stem-cell platforms to trace the evolutionary trajectories of regulatory elements, and to ask how genome organisation constrains or enables gene regulation.
